The absorbance of this complex is proportional to the albumin concentration in the sample. Contents [RGT] 4 x 100 or 1 x 1000 ml Colour reagent Citrate buffer (pH 4.2) 30 mmol/l Bromocresol green 260 µmol/l [STD] 1 x 3 ml Standard Albumin 4 g/dl or 40 g/l Sodium azide 0.095 % [STD] is standardised against the certified reference material CRM- DA470k/IFCC. Reagent Preparation [RGT] and [STD] are ready for use. Reagent Stability [RGT] and [STD] are stable up to the given expiry date when stored at 2...25°C. Contamination after opening must be avoided. Specimen Serum, heparinised or EDTA-plasma Stability in serum: at 2...8°C up to 1 month at 15...25°C up to 1 week Assay Wavelength: Hg 546 nm, 578 nm Optical path: 1 cm Temperature: 20...25°C Measurement: Against reagent blank. Only one reagent blank per series is required. Pipetting Scheme Pipette into cuvettes Reagent blank Sample or [STD] Sample/ [STD] [RGT] --- 1000 µl 10 µl 1000 µl Mix, incubate for 5 min. at 20...25°C. Measure the absorbance of the sample and the standard against the reagent blank within 30 min. ( ∆ A). Calculation of the Albumin Concentration ∆ A sample C = 4 x [ g/dl] ∆ A [STD] or ∆ A sample C = 40 x [ g/l] ∆ A [STD] Performance Characteristics Linearity The test is linear up to an albumin concentration of 6,5 g/dl or 65 g/l. Dilute samples with a higher concentration 1 + 1 with physiological saline (0.9%). Multiply the result by 2. Typical performance data can be found in the Verification Report, accessible via: www.human.de/data/gb/vr/su-albu.p Läs hela dokumentet
