Dermatophytest® è basato sulla coltura rapida su agar-agar DTM. - Raschiare la lesione con un vetrino o una lama da bisturi. - Prelevare con una pinzetta i peli periferici della lesione. Deporre peli e squame sull'agar-gel. - Umidificare dapprima l'estremità del tampone con una goccia di liquido midificatore. Passare il tampone su tutta la lesione. Strofinare il tampone sull'agar con movimenti alternati, senza schiacciare il terreno. Tempo di lettura: a partire da 2 giorni dopo la messa in coltura e a giorni alterni per 14 giorni se necessario. n CLINICAL APPLICATION
Dermatophytosis is a fungal infection of the skin encountered in
dogs, cats, rodents, and lagomorphs. It is essentially caused by
various genera of dermatophytes (fungi) and notably _Microsporum_
and
_Trichophyton._
Microscopic
examination
of
hair
and
skin
scrapings and examination under a Wood’s lamp do not provide
a definitive diagnosis. Only the growth of dermatophytes from a
sample of suspected material constitutes a final diagnosis.
Dermatophytosis is a zoonosis.
n PRINCIPLE
Dermatophytest is a selective agar medium for the diagnosis of
Dermatophytosis. When pathogenic dermatophytes of the genus
_Microsporum_ and _ Trichophyton _ are_ _ grown at room temperature,
they have the particularity to metabolize nitrogen compounds of the
medium. Thus, dermatophytes produce nitrogenous wastes which
alkalize the agar medium and lead to a color change of the agar
from yellow-orange to red in less than a week.
n PROCEDURE
1/ PREPARATION OF THE LESION:
- Wash the skin lesion with 70° alcohol or soap (non-fungicidal).
- Dry with absorbent paper.
- This eliminates any saprophytic agents that could cover the growth
of dermatophytes on the agar medium.

2/ SAMPLE:
- SKIN SCRAPE: scrape the lesion with a glass slide or the blunt side
of
a scalpel blade, from the edge of the infected area towards normal
skin. Unload hairs and scales onto the agar medium.
- HAIR pluck: deposit hairs and scales onto the agar medium.
- SWAB: moisten the extremity of a sterile swab with some drops
of moistening solution. Wipe the swab over the entire lesion and
then discharge the swab onto the agar medium using back and forth
movements, without crushing the agar medium.
