WHAT SPIRO IS USED FOR
Spiro is used:
•
To treat essential hypertension (high
blood pressure with an unknown cause)
•
To
treat
oedematous
disorders
(swelling
with
fluid),
including
congestive cardiac failure
•
For
the
diagnosis
and
treatment
of
primary
aldosteronism
(a
hormone
disorder causing fluid retention)
•
As
add-on
therapy
in
malignant
hypertension (a very serious form of high
blood pressure)
•
Where
there
is
a
low
amount
of
potassium
(a
mineral)
in
the
blood
caused
by
another
diuretic
(fluid-
removing medicine)
Spironolactone
improves
the
blood
pressure
lowering
action
of
thiazide
diuretics while at the same time reducing
or preventing potassium loss due to these
medicines.
• For the treatment of hirsutism (excess
body hair in women)
Spironolactone
is
effective
in
the
treatment
of
female
hirsutism
(excess
body
hair).
It
reduces
hair
growth,
thickness
and
hair
colour.
Increased
urine flow is unlikely to be a problem
when
Spironolactone
is
used
to
treat
hirsutism.
This
is
because
aldosterone
levels are not normally high in patients
with hirsutism.
HOW SPIRO WORKS
Spiro acts by working against a hormone
called
aldosterone.
Too
much
aldosterone causes increased amounts of
sodium
(a
mineral)
and
water
to
be
retained by the kidneys, while too much
potassium is removed from the body.
Spironolactone works against the effects
of
aldosterone.
Spironolactone
acts
by
removing excess fluid and by lowering
blood pressure. It may be given alone or
with
other
diuretics
(fluid-removing
medicines). It improves the effectiveness
of other medicines used to lower blood
pressure.
Spironolactone
also
has
a
moderate
ability to act against male sex hormones

DESCRIPTION
INDICATION
SPIRO 25MG TABLET
A 8.5 mm diameter, round, convex, marked with 'SPIRO' and scored at
one side,
plain on the other side, orange tablet. _Each tablet contains Spironolactone 25 mg._
Spironolactone is used as an adjunct in the treatment of refractory
edema, especially
when a potassium-sparing diuretic effect is desired. These may include
congestive
heart failure, hepatic cirrhosis (with or without cirrhosis),
nephritic syndrome and in
ascites associated with malignancy. It is also indicated for the
prevention and treatment
of hypokalaemia in patients for whom other measures are inappropriate
or inadequate.
It is frequently given with the thiazides,frusemide or similar
diuretics, where it adds to
their natriuretic but diminishes kaliuretic effects, hence conserving
potassium in those
at risk from hypokalaemia. It has been used as adjunctive treatment
for hypertension
with or without hyperaldosteronism, and for the diagnosis and short or
long-term
management of primary hyperaldosteronism. Other conditions in which
spironolactone
has been used on the basis of is anti-androgenic properties include
female hirsutism,
particularly in the polycystic ovary syndrome.
Spironolactone, a steroid with a structure resembling those of the
natural adrenocorticol
hormone, aldosterone, acts on the distal portion of the renal tubule
as a competitive
antagonist
of
aldosterone.
It
interferes
with
sodium
reabsorption
in
the
distal
convoluted tubule, thereby promoting excretion of sodium and water and
retention of
potassium. Spironolactone lowers blood pressure initially by reducing
plasma and
extracellular fluid volume; cardiac output also decreases. Eventually,
the extracellular
fluid
volume
and
the
cardiac
output
return
to
normal
with
an
a c c o m p a n y i n g
decrease in peripheral resistance. It also possesses antiandrogenic
effect.
Spironolactone is fairly rapidly absorbed from the gastrointestinal
tract, the bioavaila-
bility is reported to be greater than 90%. Absorption is enhanced by
